Determination of the Catalytic Activity
Concentration in Serum of
L-
Aspartate
Aminotransferase (EC 2.6.1.1 , ASAT)
BASED ON THE IFCC ASAT REFERENCE METHOD (l)
This ECCLS standard is based on the corresponding IFCC Refe–
rence Method for determi nation of ASAT, which has been adap–
ted by this ECCLS Subcommittee for routine use in European
clinical laboratories as described for CK in the General Int–
roduction to the ECCLS standards.
This standard recommends reaction conditions
a~d descr~bes
the tolerance limits for variations in measurement procedures
that nevertheless allow routine determinations of the cataly–
tic activity concentration of ASAT to be equivalent to those
of the unmodified IFCC ASAT Reference Method.
For details not given in this standard refer to the original
IFCC document, ASAT, part 2 of the IFCC methods for measure–
ment of catalytic concentration of enzymes (l).
Activation of apo-ASAT and apo-ALAT
with pyridoxal-5'-phosphate
The IFCC ASAT and ALAT Reference Methods include activation
of the apo-enzymes by pyridoxal-5'-phosphate (P-5'-P, 1 , 2).
After careful consideration of the advantages and disadvanta–
ges of this procedure the ECCLS Subcommittee decided to in–
clude this step.
